Effects of ABA and Agar on Preventing Vitrification in Carnation Plantlets Cultured in vitro



Abstract
Carnation (Dianthus caryophyllus cv. Sarinah and Scarlet Bell) cultures subcultured on 0.7% agar MS media developed into vitreous plantlets which are not transplantable to soil condition. The purpose of this study was to establish the system to obtain glaucous plantlets at the pretransplanting stage of carnation micropropagation. ABA(10-6M) supplement or/and increasing the agar concentration (over 15g/§¤) in MS medium promoted glaucous plantlet production. The MS solid medium at pH 5.5 containing 20¡­30g/§¤ sucrose resulted the highest effect of glaucous plantlet production in 16 hour/day light at 1, 700 lux of white fluorescent lamp. In the presence of several growth retardants (CCC, Ancymidol. pp-333, or B-9), the plantlet growth in vitro was inhibited, but glaucous plantlets could not be obtained. Their growth in vitro was promoted, but no glaucous plantlet production in the presence of GA©ý. Glaucous plantlets were greatly increased their survival rate(from 0 to 96%) 30 days after they had been transfered to the soil conditions.
